The overall goal of the proposed research is to determine the “stop signal” for glial-guided neuronal migration. An in vitro model system for mouse cerebellar cells will be used to analyze the role of neuron-neuron interactions in regulation of migration arrest. The specific aims are: 1. To determine whether cerebellar granule neurons terminate their migration upon encountering a lane of granule cells and/or lanes of different cell types. 2. To determine whether granule neurons terminate their migration upon encountering afferent axons from pons explants.
Significance:
Neuronal migration is a key determinant of a neuron’s final position, which in turn influences its formation of synaptic contacts and differentiation. Thus, neuronal migration is a crucial event for establishing proper structural and functional organization of the nervous system. The importance of migration to nervous system development is evident in certain mutant mice in which migration failures results in cerebellar malformation, extensive cerebellar cell death, and coordination deficits (1). Migratory abnormalities in humans are associated with a variety of neurological birth defects including congenital muscular dystrophies (2, 3). Elucidation of the mechanisms regulating migration, specifically the cues which trigger and terminate migration, is essential for understanding normal development of the central nervous system, as well as how migration abnormalities in motor cortex, cerebellum, and spinal cord might induce neuromuscular defects.
